Can the link be blocked after the purchase is refunded?

I have refunded the purchase of this download, can you block the link?

That happens automatically if the payment processor used is PayPal or Google Checkout. If you used any other payment processors (like Authorize.Net), then you can simply click on the download link a few times to expire it.
